Desert Hot Springs

You'll have several choices from DHS. The Agua Caliente in Rancho Mirage can be reached by following Dillon Road East all the way into Thousand Palms via 1000 Palms Canyon Road to Ramon Road (hang a right on Ramon). It's scenic and you'll pass right through the Coachella Valley Preserve. This route might take a while, so if you're interested in arriving as soon as possible it's probably quicker to head south on Palm Drive and hit I-10 East to Ramon.
You may also like to check out The Spa Casino in Palm Springs. For the easiest path, review the map.
Also, you can reach the Morongo Casino on I-10 heading West toward Los Angeles. In any case, it's going to take about 20-25 minutes to reach any of these locations.

Indian Wells

As with most of the casinos in the Valley, you're going to be fairly close to any number of them regardless of where you are. So, it becomes more about ease of access and in the case of Indian Wells, Fantasy Springs or Spotlight 29 are going to be the easist.
For Fantasy, simply take US Highway 111 (East) out to Indio Springs Parkway via Golf Center Parkway.

Thousand Palms

You have several great choices within 16 minutes of Thousand Palms. Your best bet is to take a few minutes to head West on Ramon and across the I-10 to the Agua Caliente. Take a left on Bob Hope and another left on Casino. 15 minutes and about 15 miles via the I-10 East you'll find Fantasy Springs Casino or another minute and another mile which includes a short second on CA-86 you'll come upon Spotlight 29.
